Laboratory determination of specific IgE antibodies

Specific IgE antibodies are determined by EIA/ELISA, RIA.
- we do not measure the concentration of IgE against pollen, we do not obtain the concentration of antibodies; we get a number that consists of two components:
- serum antibody concentration
- its binding activities,
- the more it binds, the more concentration we get: the result may be skewed,
- solid-phase change: pollen antigen is bound here, no antibody is present; if there is anti-pollen IgE in the serum, it binds to the antigen in the variable region,
- after washing the tube, add a secondary antibody labelled with an enzyme or radioisotope,
- we determine the affinity of antibodies: it is the binding through binding sites (decrease of more affine antibodies or increase of fewer affine antibodies).
